ABSTRACT

Physical exercise is a known preventive and therapeutic alternative for several cerebrovascular diseases. Therefore, the objective of the present study was to evaluate the motor performance and histomorphometry of the biceps brachii, soleus, and tibialis anterior muscles of rats submitted to a treadmill training program prior to the induction of cerebral ischemia via occlusion of the middle cerebral artery (OMCA). A total of 24 Wistar rats were distributed into four groups: Sham-Sed: sedentary control animals (n=6), who underwent sham surgery (in which OMCA did not occur); Sham+Ex: control animals exercised before the sham surgery (n=6); I-Sed: sedentary animals with cerebral ischemia (n=6); and I+Ex: animals exercised before the induction of ischemia (n=6). The physical exercise consisted of treadmill training for five weeks, 30 min/day (5 days/week), at a speed of 14 m/min. The results showed that the type-I fibers presented greater fiber area in the exercised ischemic group (I+Ex: 2347.96±202.77 µm2) compared to the other groups (Sham-Sed: 1676.46±132.21 µm2; Sham+Ex: 1647.63±191.09 µm2; I+Ex: 1566.93±185.09 µm2; P=0.0002). Our findings suggested that the angiogenesis process may have influenced muscle recovery and reduced muscle atrophy of type-I fibers in the animals that exercised before cerebral ischemia.

ABSTRACT

As neoplasias ovarianas em gatas são raras e, quando relatadas, estão associadas a animais senis, assim como a piometra de causa não iatrogênica. O objetivo deste trabalho foi relatar o caso de uma gata jovem com neoplasia ovariana, tumor de células da granulosa associado ao complexo hiperplasia endometrial cística/piometra (HECP). O animal foi atendido no setor de Reprodução Animal e Obstetrícia Veterinária (RAOV) de um Hospital Veterinário Escola, com histórico de monta natural recente, anorexia, êmese, polidipsia, poliúria e descarga vaginal purulenta. Ao exame físico, observou-se apatia, mucosas ictéricas, aumento de volume da região abdominal e presença de secreção vaginal purulenta. À ultrassonografia, foi visibilizado conteúdo ecogênico no útero diagnóstico de HECP e estrutura ecogênica na cavidade abdominal, na região ovariana, confirmado pelo exame histopatológico como sendo tumor das células da granulosa. A conduta terapêutica adotada foi a cirurgia de ovariossalpingo-histerectomia (OSH).(AU)


Ovarian neoplasias in cats are rare, and are associated with advanced ages, as well as non-iatrogenic pyometra. The objective of the present study was to report a case of a young cat showing signs of a rare neoplasia, the granulosa-cell tumor, associated with complex cystic endometrial hyperplasia/pyometra (HECP). The animal was attended in the Animal Reproduction and Veterinary Obstetrics sector of a Teaching Veterinary Hospital with a history of natural breeding, anorexia, emesis, polydipsia, polyuria, and purulent vaginal discharge. During clinical examination, apathy, icterus, swelling of the abdomen, and purulent vaginal discharge were observed. On the ultrasonographic exam, an echogenic content inside the uterus was observed, leading to diagnosis of HECP and an echogenic structure in the abdominal cavity, in ovarian region, confirmed by histopathology as being of granulosa cells. The therapeutic conduct adopted was salpingo-oophorectomy and hysterectomy surgery (OSH).(AU)

ABSTRACT

O câncer, ou neoplasia, é uma doença caracterizada pela propagação descontrolada de formas anormais das próprias células corporais e corresponde à segunda doença que mais causa mortes no mundo. A história da platina no tratamento do câncer teve início com a descoberta da sua atividade, em 1965, com a aprovação para uso clínico acontecendo apenas após 10 anos. Atualmente, os fármacos com platina estão entre os mais bem sucedidos agentes anticancerígenos, onde se destacam cisplatina (1), carboplatina (2) e oxaliplatina (3). Seus mecanismos de ação são similares: estes fármacos formam adutos com o DNA, impedindo a sua síntese e reparo, levando à morte celular. Contudo, os efeitos adversos desencadeados pelo tratamento e o desenvolvimento de resistência ao medicamento têm limitado suas aplicações. Uma das principais estratégias para a diminuição de tais efeitos consiste em alterar a estrutura destas moléculas, levando à formação de compostos híbridos, que se caracterizam pela presença de pelo menos dois fragmentos funcionais distintos em uma mesma molécula e podem apresentar maior espectro de atividade antitumoral. Dentre as alterações mais comuns encontram-se a modificação da solubilidade, através da inserção de grupos abandonadores mais ou menos hidrofóbicos e a introdução de ligantes com atividade biológica própria. Dessa forma, esta revisão visa verificar os avanços mais recentes na síntese de compostos híbridos de platina, bem como as melhorias na atividade anticâncer dos novos compostos platinados...


Cancer, or neoplasm, is a disease characterized by the uncontrolled propagation of abnormal cells of the body and is the second leading death-causing disease. The history of platinum in cancer treatment goes back to the discovery of its activity in 1965 and its approval for clinical use just 10 years later. Some of the most successful anticancer agents are Pt-based chemotherapeutics, among which cisplatin (1), carboplatin (2), and oxaliplatin (3) stand out. They have similar mechanisms of action: they form adducts with DNA, preventing its synthesis and repair and leading to cell death. However, adverse effects triggered by treatment and the development of resistance to these drugs have limited their application. One of the most important strategies to reduce such effects is to carry out structural modifications of these molecules, leading to hybrid compounds that are characterized by the presence of at least two distinct functional fragments on the same molecule and can exhibit a broader antitumor activity spectrum. Among the most typical modifications are changes to the solubility pattern, created by the insertion of leaving groups with high or low hydrophobicity, and the introduction of biologically active ligands as non-leaving groups. The purpose of these strategies is to obtain compounds capable of reducing systemic toxicity and/or overcoming acquired resistance factors to cisplatin. ABSTRACT

The objective of this study was to evaluate gastric emptying (GE) in pediatric patients with functional constipation. GE delay has been reported in adults with functional constipation. Gastric emptying studies were performed in 22 children with chronic constipation, fecal retention and fecal incontinence, while presenting fecal retention and after resuming regular bowel movements. Patients (18 boys, median age: 10 years; range: 7.2 to 12.7 years) were evaluated in a tertiary pediatric gastroenterology clinic. Gastric half-emptying time of water (reference range: 12 ± 3 min) was measured using a radionuclide technique immediately after first patient evaluation, when they presented fecal impaction (GE1), and when they achieved regular bowel movements (GE2), 12 ± 5 weeks after GE1. At study admission, 21 patients had reported dyspeptic symptoms, which were completely relieved after resuming regular bowel movements. Medians (and interquartile ranges) for GE1 and GE2 were not significantly different [27.0 (16) and 27.5 (21) min, respectively (P = 0.10)]. Delayed GE seems to be a common feature among children with chronic constipation and fecal retention. Resuming satisfactory bowel function and improvement in dyspeptic symptoms did not result in normalization of GE data.

ABSTRACT

Schistosomiasis is a major public health problem with 207 million people infected and more than 779 million at risk. The drug of choice for treating schistosomiasis is praziquantel (PZQ); however, it is inefficient against immature forms of schistosomes. The aim of this study was to test new imidazolidine derivatives LPSF/PT09 and LPSF/PT10 against adult Schistosoma mansoni worms. IC50, cytotoxicity, immune response and cell viability assays were also available for these imidazolidines. Different concentrations of imidazolidine, from 32 to 320 »M, promoted motor abnormalities in breeding and unpaired worms, and death in 24 hours at higher concentrations. Although LPSF/PT09 and LPSF/PT10 did not affect IFN-³ and IL-10 production, they induced nitric oxide production and showed a similar behavior to praziquantel on cell death test. Thus, these new imidazolidine derivatives should undergo further study to develop schistosomiasis drugs.

ABSTRACT

Small cell lung cancer (SCLC) is an aggressive disease, representing 15 percent of all cases of lung cancer, has high metastatic potential and low prognosis that urgently demands the development of novel therapeutic approaches. One of the proposed approaches has been the down-regulation of BCL2, with poorly clarified and controversial therapeutic value regarding SCLC. The use of anti-BCL2 small interfering RNA (siRNA) in SCLC has never been reported. The aim of the present study was to select and test the in vitro efficacy of anti-BCL2 siRNA sequences against the protein and mRNA levels of SCLC cells, and their effects on cytotoxicity and chemosensitization. Two anti-BCL2 siRNAs and the anti-BCL2 G3139 oligodeoxynucleotide (ODN) were evaluated in SCLC cells by the simultaneous determination of Bcl-2 and viability using a flow cytometry method recently developed by us in addition to Western blot, real-time reverse-transcription PCR, and cell growth after single and combined treatment with cisplatin. In contrast to previous reports about the use of ODN, a heterogeneous and up to 80 percent sequence-specific Bcl-2 protein knockdown was observed in the SW2, H2171 and H69 SCLC cell lines, although without significant sequence-specific reduction of cell viability, cell growth, or sensitization to cisplatin. Our results question previous data generated with antisense ODN and supporting the present concept of the therapeutic interest in BCL2 silencing per se in SCLC, and support the growing notion of the necessity of a multitargeting molecular approach for the treatment of cancer.

ABSTRACT

The objective of the present study was to evaluate changes in optic nerve head parameters, measured by confocal laser tomography, before and after trabeculectomy in order to identify outcome measures for the management of glaucoma. The optic nerve head of 22 eyes (22 patients) was analyzed by confocal laser tomography with the Heidelberg retinal tomogram (HRT) before and after trabeculectomy. The median time between the first HRT and surgery was 4.6 months (mean: 7.7 ± 8.3) and the median time between surgery and the second HRT was 10.8 months (mean: 12.0 ± 6.8). The patients were divided into two groups, i.e., those with the highest (group A) and lowest (group B) intraocular pressure (IOP) change after surgery. Differences in the 12 standard topographic parameters before and after surgery for each group were evaluated by the Wilcoxon signed rank test and the differences in these parameters between the two groups were compared by the Mann-Whitney rank sum test. Multiple regression analysis was used to evaluate the influence of the change in IOP (deltaIOP and deltaIOP percent) and the changes in the other parameters. There were significant differences in the HRT measures before and after surgery in group A only for cup volume. In group B, no parameter was statistically different. The changes in group A were not significantly different than those in group B for any parameter (P > 0.004, Bonferroni correction for multiple comparisons). deltaIOP and deltaIOP percent had a statistically significant effect on delta cup disk area, delta cup volume and delta mean cup depth. Changes in cup shape size were influenced significantly only by deltaIOP. Some optic disc parameters measured by HRT presented a significant improvement after filtering surgery, depending on the amount of IOP reduction. Long-term studies are needed to determine the usefulness of these findings as outcome measures in the management of glaucoma.

ABSTRACT

We present a critical analysis of the generalized use of the "impact factor". By means of the Kruskal-Wallis test, it was shown that it is not possible to compare distinct disciplines using the impact factor without adjustments. After assigning the median journal the value of one (1.000), the impact factor value for each journal was calculated by the rule of three. The adjusted values were homogeneous, thus permitting comparison among distinct disciplines.

ABSTRACT

Gilles de la Tourette syndrome (GLTS) is a disorder characterized by tics and several behavioral disturbances. Although GLTS is a relatively common disorder, little is known about its pathophysiology. Previous studies with SPECT and PET were performed in a small number of patients and have shown some discordant data. The aim of this study is to evaluate brain perfusion abnormalities in patients with GLTS and to correlate them with the clinical manifestations of the syndrome. Twenty-eight patients were submitted to brain [99mTc]-HMPAO SPECT. 82 percent of the patients had abnormal studies. The most frequent finding was perfusion abnormalities in the thalami in 16 patients (57 percent) and 85 percent of patients with hyperperfusion of one or both thalami had complex motor tics. This investigation has demonstrated that brain perfusion SPECT is able to identify cortical perfusion abnormalities, associated with clinical symptoms in patients with GLTS. These abnormalities involve the pre-frontal-striatal-thalamic-cortical pathways

ABSTRACT

O Plantago major é uma planta herbácea conhecida como transagem que ocorre espontaneamente nas regiões de clima temperado ou subtropical, sendo facilmente cultivada no Brasil. Popularmente é utilizada no tratamento de inflamações de boca e garganta, infecções intestinais e como agente antibacteriano. O infuso das folhas é usado como gargarejo no combate às inflamações da boca, garganta, gengivas sangrentas e parotidites. O estudo microbiológico para a avaliação da atividade do extrato hidro-alcoólico foi realizado pelo método de difusão em meio sólido, frente a doze isolados clínicos de Staphylococcus aureus, obtidos de feridas abertas da pele, secreções vaginais e da orofaringe. O extrato foi padronizado obtendo-se uma solução de concentração igual à 193,0 mg/ml. Os microrganismos foram identificados através de provas bioquímicas específicas e suas culturas foram mantidas em meio sólido Mueller-Hinton. A ciprofloxacina foi utilizada como padrão antibacteriano na concentração de 10,0 mg/ml. Os microrganismos mostraram-se sensíveis ao extrato hidro-alcoólico, apresentando halos de inibição entre 10,0 e 13,0 mm e para o padrão ciprofloxacina entre 10,0 e 15,0 mm. Baseados nos estudos acima descritos o trabalho continuará com a determinação da Concentração Mínima Inibitória (CMI).

ABSTRACT

A utilizaçäo terapêutica de doses elevadas de imunossupressores pode promover diversas complicaçöes, principalmente infecciosas. OBJETIVOS: Avaliar as complicaçöes secundárias ao uso de corticóide e ciclofosfamida em portadores de nefropatias. MÉTODOS: Foram estudados retrospectivamente 76 pacientes atendidos no Hospital das Clínicas da Faculdade de Medicina de Botucatu - UNESP, sendo divididos em três grupos: G1= Lúpus Eritematoso Sistêmico sem lesäo renal (n=15); G2= nefrite lúpica (n=33) e G3= síndrome nefrótica por glomerulopatia idiopática (n=28). RESULTADOS: Näo houve diferença em relaçäo ao tempo de acompanhamento (G1= 42,4 ± 51, G2= 52,3 ± 51, G3= 41,8 ± 47,8 meses), dose total de corticóide utilizada (G1= 20, G2= 28, G3= 16 gramas) e tempo de uso da droga (G1= 20, G2= 26, G3= 14,5 meses). Quanto ao uso de ciclofosfamida, näo houve diferença na percentagem de pacientes que a utilizaram (13 por cento no G1, 51 por cento no G2, 28 por cento no G3), porém pacientes do G1 receberam dose total menor que G2 (mediana de zero e um grama, respectivamente -- p<0.05). Aspecto cushingóide, manifestaçöes gástricas, distúrbios comportamentais, diabetes mellitus e alteraçöes oculares ocorreram nos três grupos, sem diferença estatística. Quanto às complicaçöes infecciosas, aquelas consideradas clinicamente mais graves, foram mais freqüentes no G2 (G1= 6 por cento, G2= 15 por cento, G3= 0 por cento - p<0.05), o mesmo ocorrendo em relaçäo aos óbitos (7 por cento no G1, 30 por cento no G2, 0 por cento no G3 -- p<0.05). CONCLUSÖES: Pacientes portadores de nefrite lúpica apresentaram maior freqüência de complicaçöes infecciosas decorrentes da imunossupressäo prolongada, o que pode representar um marcador de gravidade deste tipo de lesäo

ABSTRACT

O fêmur curto congênito é a deficiência longitudinal mais comum do fêmur. O membro acometido é aproximadamente 10 por cento menor do que o normal. O tratamento geralmente é feito por alongamento ósseo e as técnicas mais empregadas utilizam as corticotomias e a colocaçäo de fixador externo com alongamento progressivo. A avaliaçäo da resposta ao tratamento é difícil, baseando-se na análise radiográfica do calo ósseo em formaçäo. O objetivo deste estudo foi avaliar se a cintilografia óssea trifásica seria capaz de prever a formaçäo de calo ósseo adequado ou näo nos pacientes com fêmur curto congênito submetidos ao alongamento com fixador externo. Cinco pacientes, submetidos ao alongamento ósseo com fixador externo, foram investigados com radiografias e cintilografia óssea trifásica. Em três pacientes que apresentaram boa evoluçäo clínica a cintilografia óssea trifásica mostrou captaçäo moderada ou acentuada do radiofármaco no calo ósseo. Nos dois pacientes que apresentaram má evoluçäo clínica, a captaçäo do radiofármaco no calo ósseo foi normal ou apenas discretamente aumentada. Portanto, a captaçäo do radiofármaco na fase tardia da cintilografia óssea correlacionou-se com a evoluçäo clínica. A cintilografia óssea parece ser um método capaz de prever o prognóstico dos pacientes com fêmur curto congênito submetidos a alongamento por fixador externo

ABSTRACT

O objetivo deste estudo foi avaliar o papel da cintilografia óssea trifásica na indicação da retirada do material de síntese de pacientes com fratura diafisária do fêmur, submetidos à fixação com haste intramedular bloqueada (HIB) ou com placa em ponte (PP), já que, atualmente, a indicação da retirada é feita por critérios não totalmente confiáveis. Foram estudados 13 pacientes, seis com colocação de HIB e sete com PP. Todos os pacientes foram submetidos a cintilografia óssea trifásica com 740MBq de MDP-99mTc em uma câmara de cintilação computadorizada. Em quatro pacientes (dois com HIB e dois com PP) houve retirada do material de síntese, em média, 42,2 meses após a fratura. Os pacientes com HIB apresentaram alterações discretas na fase tardia da cintilografia óssea e formação discreta do calo ósseo, enquanto os com PP mostraram alterações cintilográficas moderadas e formação anormal do calo ósseo. As discretas alterações descritas na cintilografia óssea nos pacientes com HlB indicam que este tipo de fixação leva a menor exigência mecânica do calo ósseo com remodelação mais fisiológica, comprovada pelo estudo tomográfico em dois pacientes, mostrando sua anatomia semelhante à do fêmur contralateral. Isto não ocorreu nos pacientes com PP, talvez devido a uma maior movimentação dos fragmentos ósseos permitida por este material. Esta movimentação sugere que este tipo de fixação talvez não seja o mais fisiológico, não proporcionando à fratura condições adequadas para se consolidar.

ABSTRACT

Recentemente a core biopsy estereotática tem sido proposta como método alternativo na avaliação inicial de lesões mamárias impalpáveis. Objetivo. Os autores propõem-se a analisar a acurácia do diagnóstico histopatológico em material de core biopsy estereotática. Discutem a aplicabilidade de testes usados para variáveis nominais como sensibilidade e especificidade na avaliação de gradações de qualidade, tais como, benigno, borderline e maligno. Material e Métodos. De um total de 700 biópsias, selecionou-se para o estudo as 567 primeiras (obtidas até maio de 1996), com acompanhamento de no mínimo seis e no máximo trinta meses. Foi feita a comparação do diagnóstico dado na core com achados da peça cirúrgica ou do acompanhamento clínico-mamográfrico subseqüente. Resultados. A sensibilidade foi de 90,72 por cento e a especificidade de 98,36 por cento. O teste de correlação de Spearman foi 0,92 (máximo 1). Conclusão. Os dados são semelhantes aos da literatura e demonstram uma boa acurácia da core biopsy no diagnóstico histopatológico de lesões impalpáveis da mama.

ABSTRACT

The identity of the histamine-potentiating activity detected in the rat anaphylactic pleural washing was investigated. Wistar rats of both sexes, weighing 150-200 g, were sensitized by injecting subcutaneously (sc) a mixture of ovalbumin and Al(OH)3 14 days before allergen challenge. In sensitized rats, intrapleural (ipl) injection of ovalbumin (l2 mug/cavity) caused an intense protein exudation. A single ipl administration of compound 48/80 (l2 mug/cavity) exhausted the resident mast cell population and turned the pleural cavity hyporeactive to the allergen challenge performed 5 days later. Allergen-induced exudation occurred in parallel to a dramatic decrease in the amount of cell-stored histamine (from 9.6 ñ 1.4 (N = 8) to 1.3 ñ 0.1 (N = 6) mug/cavity, P<0.001) in the pleural fluid within 10 min. The anaphylactic cell-free pleural washing obtained at this time, as well as histamine at a concentration equivalent to that stored in pleural mast cells (10 mug/cavity), did not induce pleural exudation when injected into normal rats. In contrast the combined administration of histamine and anaphylactic pleural washing led to remarkable pleural exudation, comparable to that obtained with a high dose of histamine (200 mug/cavity) alone. It is noteworthy that the anaphylactic washing from compound 48/80 pretreated rats failed to synergize with histamine. Also, synergism was not reproduced when recipient rats were pretreated with methysergide (50 mug/cavity). Consistently, serotonin (5 mug/cavity) acted synergistically with histamine (1O mug/cavity), producing a greater exudative response than observed with the sum of the effects of each vasoactive amine alone. The results indicate that serotonin accounts for the histamine-potentiating activity noted in the anaphylactic pleural washing, confirming that the synergistic interaction between these vasoactive amines plays a critical role in the rat allergic pleurisy.

ABSTRACT

A ultra-sonografia do tipo B (SCANNER 450 (5MHz), Pie Medical, Holanda) foi utilizada isolada ou em conjunto com exames laboratoriais e radiográficos em 33 cadelas com história clínica compatível com piometra. O útero dilatado apresentou imagem ultra-sonográfica de uma estrutura tubular bem definida com diâmetro entre 0,5 e 4,0 cm. O conteúdo luminal uterino apresentou menor ecogenicidade que a parede, com cintilaçöes ecogênicas bem evidentes. Houve concordância entre o aumento da viscosidade da secreçäo e a intensidade ecogênica. O diagnóstico ultra-sonográfico foi possível em 31 animais (94 por cento), e foi confirmado através de laparotomia ou necrópsia. Concluímos que a ultra-sonografia do tipo B é método eficiente, podendo ser utilizado no diagnóstico da piometra canina

ABSTRACT

This study was undertaken to characteize the different phases of the allergic pleurisy induced by ovalbumin in actively sinsitized rats. The reaction was triggered by the intrathoracic injection of ovalbumin (12 microng/cavity) into animals sensitized 14 days before. The challenge caused, at 30 mjin, a drastic mast cell degranulation and exudation which peaked within 4h. At this time, an intense pleural leucocyte recruitment also occurred, accounted for by an increase in the mononuclear cell counts and by a predominant influyx of neutrophils. After 24h, the mast cell counts stated to reover, accompanied by a long-lasting (96 h) accumaltion of pleural eosinophils. Forty-eight hour later, the exudation and neutrophils were at basal levels, whereas mast cell counts increased progressively to reach control values at 120 h. This study describes the time course of the exudatory and cellular alterations observed during pleural inflammation induced by low antigen concentrations
